The three guards combined for 65 points as the Golden Eagles beat the Pirates 92-69.
Southern Miss(11-10, 3-4 Conference USA) has won two of its last three games.
The Pirates (7-12, 1-5) have lost four straight and have dropped 20 straight league games on the road.
Wise scored a game high 28 points, including 19 in the first half when Southern Miss took control of the game for good.
"We came out strong in the first half and played well, "Wise said. "We were shooting the ball well and that seemed to get everyone into the game."
Southern Miss broke the game open for good with a 13-1 run midway through the first half. Wise and Horton each had 3-pointer's in the run, which turned a 16-16 tie into a 29-17 lead.
"It was a very good effort by our guys," Southern Miss coach Larry Eustachy said. "When the defense can trigger some easy shots at the basket then that usually helps you shoot the ball better."
Southern Miss took advantage of cold shooting by the Pirates and some sloppy ball-handling in the first half. The Pirates hit just 29 percent from the field and had 11 turnovers and trailed 55-30 at halftime.
"We have to find a way to win a game on the road in this league," East Carolina head coach Mack McCarthy said. "And you won't do that playing like we did tonight, especially in the first half."
The Golden Eagles led by as much as 33 points in the second half.
Horton added 22 points for Southern Miss and Courtney Beasley chipped in 15. Sai'Quon Stone had 10 rebounds for the Golden Eagles.
